Sample collection
Preparation and processing of swab pools for detection of Influenza A matrix gene and/or Avian Orthoavulavirus Type 1 by real time RT-PCR
This protocol details the extraction of Avian Influenza A or avian paramyxovirus type 1 (APMV-1) viral RNA for rapid detection from pooled swabs with the aim of receiving an early warning for the presence of influenza A virus and AOAV-1.
Pooled swabs can consist of up to 5 cloacal or 5 oropharyngeal swabs. Following RNA extraction, PCR can be used to amplify the nucleic acid.
This method is applicable to statutory testing for notifiable avian disease (NAD) including International Trade, report cases, surveillance testing following the detection of NAD in poultry or wild birds, and NAD exclusion testing.
Procedure for extraction of samples from avian carcasses for virus detection and isolation
This protocol details the collection of samples from avian carcasses which have been submitted for detection of avian paramyxovirus type 1 (APMV-1) and Influenza virus infection.
Tissue samples are taken from a variety of organs. The collected samples can be used for procedures including virus isolation in eggs, virus isolation in cell culture, extraction for virus detection and pathological examination of avian carcasses.
